The Four Layers of Skin Hydration

Layer 1: The Stratum Corneum & TEWL

The outermost skin layer is a "brick and mortar" structure of corneocytes embedded in a lipid matrix of ceramides, cholesterol, and fatty acids. Its primary function is preventing transepidermal water loss (TEWL). Layer 2: The Natural Moisturizing Factor (NMF)

The NMF is a collection of water-soluble compounds within corneocytes that bind and retain water — amino acids (40%), pyrrolidone carboxylic acid (12%), lactate (12%), and urea (7%). Layer 3: Aquaporins (Cellular Water Channels)

Aquaporin-3 (AQP3) is the primary water channel in the epidermis — transporting water and glycerol into keratinocytes. AQP3 expression declines with age and UV exposure. Niacinamide and glycerol upregulate AQP3 expression, improving cellular water transport. (Hara et al., PMID: 12100180)

Layer 4: The Dermal Matrix & Hyaluronic Acid

The dermis contains hyaluronic acid (HA) that binds up to 1,000x its weight in water, maintaining the plumpness and volume that gives skin its youthful appearance. Dermal HA declines ~50% between ages 20 and 60 (Longas et al., PMID: 15304189) — producing volume loss and static wrinkles that no surface moisturizer can address.

Hyaluronic Acid Molecular Weight: Why It Matters

Not all hyaluronic acid is equal — molecular weight determines where HA acts in the skin:

High MW HA (>1,000 kDa)

Surface film formation. Reduces TEWL. Immediate plumping. Does not penetrate.

Medium MW HA (100–500 kDa)

Penetrates upper epidermis. Hydrates stratum corneum from within. Supports NMF.

Low MW HA (<50 kDa)

Hyaluronic Acid 🟢 Strong Evidence

Kawada et al. (2014, PMID: 25014997) demonstrated oral HA (120mg/day) over 12 weeks significantly improved skin moisture and reduced wrinkle area vs placebo. Niacinamide (Vitamin B3) 🟢 Strong Evidence
