Zinc is a cofactor in over 300 enzymes and a structural component of over 1,000 transcription factors. Its skin-specific roles are remarkable: zinc is essential for collagen synthesis (zinc-dependent prolyl hydroxylase), wound healing (zinc finger proteins in growth factor signalling), sebum regulation (inhibits 5Ξ±-reductase and reduces DHT-driven sebocyte stimulation), anti-inflammatory activity (inhibits NF-ΞΊB and reduces IL-1Ξ², IL-6), and antioxidant defence (zinc-superoxide dismutase, or Zn-SOD). Zinc deficiency β present in an estimated 17% of the global population and significantly higher in plant-based eaters β produces a characteristic dermatological picture: acrodermatitis, impaired wound healing, hair loss, and increased inflammatory skin conditions.
Zinc & Acne: Better Evidence Than You Think
Oral zinc is one of the most evidence-supported non-prescription interventions for acne vulgaris. Multiple RCTs demonstrate oral zinc (zinc gluconate 30mg or zinc sulfate 220mg daily) reduces inflammatory acne lesion count by 30β60% β comparable to low-dose doxycycline in some studies, without antibiotic resistance risk. Mechanisms: zinc inhibits 5Ξ±-reductase (reducing DHT-driven sebum), suppresses C. acnes growth (zinc is bacteriostatic), reduces keratinocyte IL-1Ξ² and TNF-Ξ± production (anti-inflammatory), and inhibits MMP activity in acne lesions (Gupta M et al., 2014 β PMID: 24719062). Topical zinc (zinc pyrithione, zinc PCA) has additional direct sebostatic and antibacterial effects at the skin surface.
Zinc & Wound Healing
Zinc is essential at every phase of wound healing: haemostasis (zinc-dependent platelet aggregation), inflammation (zinc-finger transcription factors regulate cytokine expression), proliferation (zinc-dependent collagen hydroxylation and cross-linking), and remodelling (zinc-MMP regulation). Zinc deficiency significantly impairs wound healing speed and quality. In surgical and chronic wound studies, zinc supplementation in deficient patients accelerates re-epithelialisation and increases wound tensile strength. Topical zinc oxide creates a physical barrier, has mild antimicrobial properties, and reduces inflammation at wound sites.
Zinc & Anti-Aging: The Overlooked Connection
Zincβs anti-aging skin relevance operates through three distinct mechanisms. First, Zn-SOD (zinc-superoxide dismutase) is the primary intracellular superoxide scavenger β zinc deficiency reduces Zn-SOD activity and increases oxidative stress-driven collagen degradation. β οΈ Honest Limitations
Zinc supplementation in zinc-sufficient individuals has limited additional benefit. The impressive acne and wound healing data comes primarily from studies in zinc-deficient populations. If you are already zinc-replete, additional supplementation adds little. Test before supplementing (serum zinc, though imperfect, is accessible).
High-dose zinc (>40mg elemental zinc daily) depletes copper. Zinc and copper compete for absorption. Long-term high-dose zinc supplementation without copper co-supplementation causes copper deficiency β which paradoxically increases cardiovascular risk. Use zinc at physiological doses (8β15mg elemental) for daily maintenance.
